In vitro drug release profile moringa oleifera seeds extract loaded chitosan nanoparticles (MOSE-CNP) via dialysis and spectrophotrophotometric analysis

Abstract
Background: Natural product-based drug delivery systems have been developed as a promising strategy to improve the bioavailability of therapeutic molecules. Aim: This research describes the development and characterisation of Moringa oleifera seeds extract (MOSE) loaded chitosan nanoparticles (MOSE-CNPs) as an innovative carrier for bioactive compound. Methods: The MOSE-CNPs were prepared via ionic gelation method with thorough investigation by different techniques such as scanning electron microscopy (SEM), dynamic light scattering (DLS), FTIR, and UV–vis spectroscopy. The in vitro drug release profile was characterized by a dialysis membrane method in phosphate buffered saline (PBS) of pH 7.4 for up to 72 h. Results: The average particle size of the fabricated nanoparticle was 185.4 ± 12.6 nm, PDI was 0.289 ± 0.031 and EE were 78.3 ± 3.2%. The release profile of the nanoparticles was controlled and sustained with approximately 35.6% drug released in the first day, and around 72.8% cumulative release within 72 hours. The zeta potential analysis revealed the positive charge of +42.3 ± 2.1 mV, which provided the electrostatic stability to the formulation. Conclusion: The results suggest that MOSE-CNPs can serve as an efficient delivery system to improve the oral bioavailability and therapeutic effect of natural bioactive agents

المقدمة:طُوِّرت أنظمة توصيل الأدوية القائمة على المنتجات الطبيعية كاستراتيجية واعدة لتحسين التوافر الحيوي للجزيئات العلاجية. الهدف: يصف هذا البحث تطوير وتوصيف جسيمات نانوية من الكيتوزان محملة ب مستخلص بذور المورينجا أوليفيرا (MOSE) كناقل مبتكر للمركبات النشطة بيولوجيًا. طرق العمل: حُضِّرت جسيمات MOSE-CNPs النانوية باستخدام طريقة التجليد الأيوني، مع دراسة شاملة باستخدام تقنيات مختلفة مثل المجهر الإلكتروني الماسح (SEM)، وتشتت الضوء الديناميكي (DLS)، ومطيافية الأشعة تحت الحمراء بتحويل فورييه (FTIR)، ومطيافية الأشعة فوق البنفسجية والمرئية (UV-vis). وتم توصيف خصائص إطلاق الدواء في المختبر باستخدام طريقة غشاء الديلزه في محلول ملحي فوسفاتي (PBS) عند درجة حموضة 7.4 لمدة تصل إلى 72 ساعة. النتائج: بلغ متوسط حجم الجسيمات النانوية المصنعة 185.4 ± 12.6 نانومتر، وبلغ مؤشر تشتت الحجم 0.289 ± 0.031، وكفاءة التغليف 78.3 ± 3.2%. وتم التحكم في معدل إطلاق الدواء من الجسيمات النانوية واستدامته، حيث تم إطلاق حوالي 35.6% من الدواء في اليوم الأول، وحوالي 72.8% من الإطلاق التراكمي خلال 72 ساعة. وكشف تحليل جهد زيتا عن شحنة موجبة قدرها +42.3 ± 2.1 ملي فولت، مما وفر الاستقرار الكهروستاتيكي للتركيبة. الاستنتاج: وتشير النتائج إلى أن جسيمات الكيتوسان النانوية المحملة بـ MOSE يمكن أن تُستخدم كنظام توصيل فعال لتحسين التوافر الحيوي عن طريق الفم والتأثير العلاجي للعوامل النشطة بيولوجيًا الطبيعية.
